First proposed in the House of Burgesses in 1767, Botetourt County was created in 1770 from Augusta County. The county is named for Norborne Berkeley, 4th Baron Botetourt, who served as governor of the colony of Virginia from 1768 to 1770, when he died suddenly while in office. In 1772, the county was reduced to the area east of the New and Kanawha rivers by the creation of Fincastle County. Most of that latter county became the Commonwealth of Kentucky in 1792. The other counties established directly from portions of Botetourt County are: Rockbridge (1778), Bath (1791), Alleghany (1822), Roanoke (1833), and Craig (1851).

The town of Fincastle was originally settled by Scotch-Irish and German immigrants from Pennsylvania who arrived in the region in the mid-18th century. John Miller erected a sawmill which became the nucleus of the early village of Fincastle. In 1770, Botetourt County was separated from Augusta County, and Miller's home was selected as the meeting place for the court. Two years later, Israel Christian donated 45 acres (18 ha) of land and laid out the streets and lots for the new town of Fincastle. The town's name was selected to honor George, Lord Fincastle, the eldest son of Virginia's Lieutenant Governor, Lord Dunmore. In 1773, the first log courthouse was built on the designated court square at the center of Fincastle. By the early-19th century, the village had grown into an important regional center of commerce, especially for the wool industry, aided by the construction of taverns, hotels, and shops.

Buchanan was incorporated in 1832. The town of Pattonsburg was founded on the opposite side of the James River (named for Colonel James Patton), and was connected to Buchanan via a bridge. An 1855 gazetteer described Buchanan and Pattonsburg together containing "3 or 4 churches, 1 bank, 1 printing office, and several tobacco factories and mills." Buchanan was named for Virginia colonist John Buchanan (died 1769), a soldier and landowner who built Fort Fauquier on the James River, just south of the town.

The railroad reached the Troutville area in 1881, and a station was built, but the area had no name. As three nearby houses were occupied by the brothers John, Jim, and Jorge Trout, the area was called Troutsville. It became an important shipping center in the 1890s. In 1895, Troutsville was a thriving town with an active shipping point, farming, canneries, packing houses and factories. The name of the town was later changed to Troutville. The town was chartered on June 4, 1956.
